What does Mars rule for a Virgo ascendant?
Mars rules the 8th and 3rd houses for you, because Mars owns Aries and Scorpio and those signs fall in those positions counted from Virgo.
Mars rules your 3rd and 8th houses. The 8th rulership is worth noting because it makes Mars a functional malefic of a particular kind for you, and Virgo cannot access the sign-house exception at all.
Is Mars Yogakaraka for a Virgo ascendant?
No. Mars is Yogakaraka only for Cancer and Leo ascendants, where it rules both an angle and a trine. For Virgo it rules the 8th and 3rd houses, which does not meet that condition.
This matters practically: rule 7 is worth 30 points, the second-strongest of the ten, and it is simply unavailable to you. Cancellation has to come from elsewhere.

Which trigger placement is least severe for you?
Aries in the 8th house — because Mars carries a dignity there that triggers a cancellation rule in its own right. A dosha registered from one of those placements arrives with 25 points of cancellation already attached, before Jupiter, the Navamsa or anything else is considered.

Can rule 8 apply to a Virgo ascendant?
No. Rule 8 is structurally unavailable to a Virgo ascendant, and no chart can change that.
The five named exceptions are Aries in the 1st, Scorpio in the 4th, Capricorn in the 7th, Cancer in the 8th and Sagittarius in the 12th. Each of those sign-and-house pairs occurs from exactly one ascendant — Aries, Leo, Cancer, Sagittarius and Capricorn respectively. Virgo is not among them.
This is worth knowing rather than discovering by accident. The strongest cancellation rule in the method, worth 35 points, is simply not on your table, and any calculator that reports it for you has made an error.
